Synthesis of Cu(II), Ni(II), Zn(II), Co(III) and Fe(III) complexes using Deferoxamine and their DNA binding affinity, plasmid DNA cleavage studies

New and versatile route for the preparation of Deferoxamine from 5-aminopentan-1-ol and their Cu(II), Ni(II), Zn(II), Co(III) and Fe(III) (11 to 15) complexes were synthesized and characterized. The mode of binding interactions of synthesized complexes 11 to 15 with CT – DNA was monitored by absorption spectral titrations, fluorescent spectral titrations and viscosity measurements. The intrinsic binding constant of 11 to 15 was determined from absorption spectral data of increasing amounts of CT-DNA into aqueous solution of complex (10 μM). An aqueous solution of 11 to 14 complexes cleaves the pBR 322 DNA. The complex 11 damages the plasmid DNA by oxidative cleavage and hydroxyl radical plays vital role in DNA damage. 12 to 14 cleave the plasmid DNA by hydrolytic pathway. Gel electrophoresis using groove recognition agents DAPI (minor groove binder) and methyl green (major groove binder) has shown that complexes bind in the minor groove of DNA. © 2013 Universal Research Publications.
